      目的:观察二陈汤对非酒精性脂肪性肝病(non-alcoholic fatty liver disease, NAFLD)法尼酯衍生物X受体(farnesoid X receptor, FXR)信号通路及胆汁酸水平的影响。 方法:以高脂饮食诱导NAFLD模型,以二陈汤高、中、低剂量干预,以奥贝胆酸为阳性对照。干预4周后对肝脏组织进行HE染色,对肝脏组织及血清胆汁酸水平进行测定,并以Realtime-PCR法和Western blot法检测FXR、SHP、CYP7A1和BSEP mRNA及蛋白表达水平。 结果:二陈汤不同剂量及奥贝胆酸均可不同程度地改善NAFLD肝脏组织病理学形态、降低肝脏及血清胆汁酸水平、上调肝脏FXR、SHP、BSEP,下调CYP7A1 mRNA和蛋白表达,且以二陈汤高剂量疗效最佳。 结论:二陈汤可激活NAFLD大鼠FXR信号通路,降低胆汁酸水平,改善肝脏脂肪病变。

      Objective. To observe the effect of Erchen Decoction on FXR signal pathway and bile acid level in non-alcoholic fatty liver disease. Methods. The NAFLD rat model was induced by high-fat diet. The trial group received three different dosages of Erchen Decoction. Obeticholic acid was chosen as the positive control drug. After 4 weeks treatment period, liver tissues HE staining was performed. Meanwhile, the liver tissue and serum bile acid levels were measured. The mRNA and protein expression levels of FXR, SHP, CYP7A1 and BSEP were evaluated by Realtime-PCR and Western blot respectively. Results. Both Erchen Decoction and obeticholic acid showed the effect of improving the histopathological morphology of liver, decreasing the liver and serum bile acid levels, up regulating the mRNA and protein expressions of FXR, SHP, BSEP, and down regulating CYP7A1. Additionally, the high dosage of Erchen Decoction showed the most superior effect. Conclusion. Erchen Decoction could active the FXR signaling pathway in NAFLD rats, then reduce the bile acid level, improve the histological features of liver.
