      目的:研究炙甘草汤对阿霉素诱导大鼠心力衰竭能量代谢的影响。方法:预防组及防治结合组于造模前2周开始灌胃炙甘草汤(10ml/kg),每日2次,第3周开始分别在模型组、预防组、治疗组和防治结合组尾静脉注射阿霉素(ADM)4mg/kg,5天一次,给药3次,建立大鼠心肌损害模型。造模成功后,治疗组及防治结合组在尾静脉注射当天,继续予炙甘草汤灌胃,对照组以相应体积的生理盐水灌胃。以高效液相色谱法检测各组大鼠心肌组织中心肌磷酸腺苷(ADP、ATP)的表达水平,酶联免疫吸附(ELISA)测定各组血清心肌肌钙蛋白(cTnI)水平。结果:与正常组相比模型组cTnI含量显著升高(P<0.01);防治结合组与模型组相比呈下降趋势(P<0.05);预防组、治疗组与防治结合组均显著下降(P<0.01)。模型组ATP和ADP浓度均下降(P<0.05),预防组与对照组相比ATP浓度降低(P<0.05),但是与模型组相比升高(P<0.05)。结论:炙甘草汤有效调节心衰大鼠的心肌能量代谢,延缓了心衰的发生,效果明显。

      objective: To study the effect of Zhigancao decoction on energy metabolism of Adriamycin induced heart failure in rats. Methods: Two weeks before the establishment of the model, the prophylaxis group and the combined prophylaxis-treatment group were administered Zhigancao decoction orally (10ml/kg), twice a day. At the beginning of the 3rd week, Adriamycin (ADM) was injected into the tail vein of the model groups, the prevention group, the treatment group and the combined prevention-treatment group at 4mg/kg, once every 5 days, and three times, respectively, to establish the rat myocardial damage model. After the successful establishment of the model, the treatment group and the combined prevention-treatment group were given Zhigancao decoction by gavage on the day of the injection of tail vein, and the control group was given normal saline solution with corresponding volume. The expression levels of adenosine phosphate (ADP, ATP) in the myocardial tissues of rats in each group were determined by high performance liquid chromatography (HPLC), and the serum levels of cardiac troponin (cTnI) in each group were determined by enzyme-linked immunosorbent assay (ELISA). Results: Compared with the normal group, the content of cTnI in the model group was significantly increased (P<0.01). Compared with the model group, the combined prevention-treatment group showed a downward trend (P<0.05). The prevention group, the treatment group and the combined prevention-treatment group decreased significantly (P<0.01).The concentration of ATP and ADP in the model group decreased (P < 0.05), and that in the prevention group decreased evidently (P < 0.05) with the comparison of the control group, but increased (P < 0.05) with the comparison of the model group. Conclusion: Zhigancao decoction can effectively regulate the myocardial energy metabolism of rats with heart failure and delay the occurrence of heart failure.
