      摘要:目的:研究生脉饮联合吉西他滨对膀胱癌T24细胞增殖、迁移、侵袭、凋亡, 及荷瘤小鼠瘤体生长的影响。方法:将膀胱癌细胞株T24随机分成4组,分别为空白组、生脉饮组、吉西他滨组、联合用药组。药物干预T24细胞24、48、72h后,采用CCK-8法检测T24细胞增殖活性;Transwell法观察细胞迁移、侵袭的情况;流式细胞术检测生脉饮及联合吉西他滨对T24凋亡的影响;采用右前腋下接种膀胱癌T24细胞的方法构建膀胱癌荷瘤小鼠模型,评价生脉饮联合吉西他滨对小鼠体内肿瘤重量与体积的影响。结果:与空白组相比,生脉饮组T24细胞增殖、迁移与侵袭能力趋于减弱且凋亡有所增强,而两者无统计学差异 (P>0.05);此外,联合用药组T24细胞增殖、迁移与侵袭能力显著降低,细胞凋亡率明显增加,且小鼠肿瘤重量与体积均显著降低 (P<0.05)。结论:生脉饮一定程度上抑制膀胱癌T24细胞的生长,且有助于增强吉西他滨对膀胱癌的抑制效应。

      Abstract: Objective To explore the effects of shengmai decoction combined with gemcitabine on the proliferation, migration, invasion, apoptosis of bladder cancer T24 cells and tumor growth in tumor-bearing mice. Methods The bladder cancer cell line T24 cells were randomly divided into 4 groups: blank group, shengmai decoction group, gemcitabine group, and combination group. After 24, 48 and 72h of drug intervention, CCK-8 was used to measure the proliferation of T24 cells. Transwell was used to detect the invasion and migration of T24 cells. The apoptosis of T24 cells that treated with shengmai decoction alone and its combination with gemcitabine was detected by flow cytometry. The bladder cancer tumor-bearing mice model was constructed by inoculating bladder cancer cells T24 in the right anterior armpit for assessing the tumor weight and volume of mice with treatment of shengmai decoction alone and its combination with gemcitabine. Result Compared with the blank group, the proliferation, migration and invasion ability was slightly attenuated, and the apoptosis rate has increased in shengmai decoction group, but there was no statistical difference (P>0.05). Additionally, the proliferation, migration and invasion of T24 cells were significantly decreased, and the apoptosis rate was observably increased in combination group, as well as the tumor weight and volume were significantly reduced (P<0.05). Conclusion Shengmai decoction treatment inhibits the growth of bladder cancer T24 cells to a certain extent, and could enhance the inhibition of gemcitabine in bladder cancer.
