目的:建立肺宁合剂传统饮片汤剂与中药配方颗粒汤剂中麻黄碱、伪麻黄碱、苦杏仁苷、甘草苷、甘草酸、白花前胡甲素、白花前胡乙素7种指标成分HPLC-UV分析方法,并对二者所含的多指标成分含量进行分析评价,为中药配方颗粒的进一步研究与应用以及肺宁合剂质量标准提升提供思路和参考。方法:采用HPLC-UV同时测定肺宁合剂传统饮片汤剂与中药配方颗粒汤剂中7种指标成分。应用Thermo-C18 endcapped 色谱柱(250 mm×4.6 mm, 5 µm),以0.1%磷酸水-乙腈为流动相梯度洗脱,柱温30 ℃,检测波长210 nm,流速0.8 mL/min,进样量10 μL。结果:所建立的方法线性关系良好,精密度、稳定性、重复性、加样回收率均符合液相含量测定的要求。肺宁合剂传统饮片汤剂与中药配方颗粒汤剂中均含有7种指标成分,但肺宁合剂传统饮片汤剂中的麻黄碱、伪麻黄碱和甘草苷含量显著高于中药配方颗粒汤剂,而苦杏仁苷、甘草酸、白花前胡甲素和白花前胡乙素含量无明显差异。结论:中药配方颗粒是否可以替代传统饮片生产的肺宁合剂值得进一步研究,应进一步完善中药配方颗粒提取工艺及质量标准以适应临床应用,研究结果亦可为肺宁合剂质量标准的提升提供参考。 |

Objective: To establish a HPLC-UV method for the determination of ephedrine, pseudoephedrine, amygdalin, glycyrrhizin, glycyrrhizic acid, imperatorin A and imperatorin B in the traditional decoction of Feining mixture and granule Decoction of Chinese medicinal granules, and to analyze and evaluate the contents of the multi-index components in the two decoctions for the further research and application of Chinese medicinal granules as well as the improvement of quality standards of Feining mixture. Method: Simultaneous determination of seven index components in traditional decoction of Feining mixture and granule Decoction of Chinese medicinal granules by HPLC-UV was carried out. Thermo-C18 endcapped column (250 mm×4.6 mm, 5 µm) was used for gradient elution with 0.1% phosphoric acid-acetonitrile as mobile phase. The column temperature was 30℃, the detection wavelength was 210 nm, the flow rate was 0.8 mL/min, and the injection volume was 10μL. Result: The established method has a good linear relationship, and its precision, stability, repeatability and recovery all meet the requirements of liquid phase content determination. The traditional decoction of Feining mixture and granule Decoction of Chinese medicinal granules contain seven index components, but the contents of ephedrine, pseudoephedrine and glycyrrhizin in traditional decoction of Feining mixture are significantly higher than those in granule Decoction of Chinese medicinal granules, while the contents of amygdalin, glycyrrhizic acid, prehualin A and prehuacin B are not significantly different. Conclusion: Whether the Chinese medicinal granules can replace the Feining mixture deserves further study. It is necessary to further improve the extraction technology and quality standard of the Chinese medicinal granules to adapt to clinical application. The research results can also provide reference for the improvement of the quality of Feining mixture. |
